God provides Essay

SEEKING GUIDANCE The discipline of seeking guidance goes to the heart of walking with God. Guidance involves us hearing God’s voice and obeying Him. It is not only seeking His will in a specific matter, but it is a constant way of life. God provides guidance to His people in two ways. One is individual guidance, where God gives each of His children divine instruction through Scripture, reason, circumstances, and through prompting of the Spirit in the individual’s heart. In corporate guidance, God guides groups of people who gather in fellowship to seek the will of God. Both aspects are important to submit to as we seek to live in response to the voice of God. It is also a treatise on the nature of the perception of God and spiritual growth. One must believe in divine guidance. One must believe God will guide you. What God wants from each individual. Why each individual needs to seek God’s guidance. Most often, God sends divine guidance and wisdom through anointed people and not supernatural signs. You need wise counsel. Wisdom is usually gained by successful experience. Everyone has experience but only a few have successful experience. Many live in a continual cycle of action then regret but wisdom either successfully breaks bad cycles or never gets in bad cycles. It is usually far less painful to benefit from the experience of others than having to go through it ourselves. When you successfully go through something, you usually have wisdom. Everybody Knows: You can’t be all things to all people. You can’t do all things at once. You can’t do all things equally well. You can’t do all things better than everyone else. Your humanity is showing just like everyone else’s. Therefore:You have to find out who you are, and be that. You have to decide what comes first, and do that. You have to discover your strengths, and use them. You have to learn not to compete with others, Because no one else is in the contest of being you. Then: You will have learned to accept your own uniqueness. You will have learned to set priorities and make decisions. You will have learned to live with your limitations. You will have learned to give yourself the respect that is due. And you’ll be a most vital mortal. Dare To Believe: That you are a wonderful, unique person. That you are a once-in-all-history event. That it’s more than a right, it’s your duty, to be who you are. That life is not a problem to solve, but a gift to cherish. And you’ll be able to stay one up on what used to get you down. Loving ourselves being as compassionate, gentle & loving with ourselves as we would with a best friend can be pretty hard to do. This is especially true for women & survivors of abuse (including neglect). As women & survivors, we’ve been trained to deny our own feelings & needs & to take care of others. And we’re also frequently given messages that tell us not to accept or love ourselves. This is especially true for survivors; it’s so easy for us to take in the hating messages our abusers gave us & to turn that inwards on ourselves. But it is possible to love ourselves or at least to increase our self-love in increments, until we can know, deep to our cores, that we love ourselves & that we’re beautiful. Here are some of the things that have worked for me. I hope you’ll find they work for you, too. Carrying around some form of comfort & having it available right when you need it can help ease anxiety & stress & give you reassurance when you need it. I’ve found that just knowing I have a comfort bag with me is sometimes enough – and other times, I am grateful I have it with me to dip into. You might want to try carrying around some comfort & see if it makes a difference in your life. There are times when we are out in the world & need a little – or a lot – of extra comfort. Times when we are nervous or scared, feeling vulnerable or unsure of ourselves, or just not very confident. A new job, a speech we have to give, a new situation, a group where we don’t feel very welcome, or just when you’re feeling stressed out. Or, for survivors, facing something triggering, frightening, or painful. It’s times like those that portable comfort can come in handy. Portable comfort means bringing something with you on purpose that helps you to feel comforted & reminds you that you are safe & loved. There are many different ways of bringing comfort with you – and they don’t all have to be obvious. I take portable comfort with me whenever I leave the house. I have things already placed in my backpack, the pockets of my coat & sometimes even my jeans, so if I forget to bring something extra with me, I already have something with me. And I always wear the same necklace; it’s always with me. If you have time before you leave for the day, it’s a good idea to associate whatever good feeling you want to remind yourself of (comfort, safety, love) with the object. To do this, take a moment & remember when you felt comforted, safe, or loved. Hold that feeling to you. Now imagine that good feeling flowing into the object. Tell yourself that whenever you see or feel or notice that object in your day, you will be reminded of that good feeling & feel it again.

African Cinema

African cinema is inaccurate to be tagged as a â€Å"national cinema† for Africa has more than 50 states. The interaction of these nations with European countries, with the exception of some parts of the South Africa and other Mediterranean countries like Morocco, Egypt, Algeria, and Tunisia, paved for the development of film-making (Abrams, Bell, and Udris, 2001).In fact, not until 1980s, the government of the African nations failed to show compassion for the film-making which resulted to the inability to produce film that is indigenous to their own culture. The repeated app earance of these objects in the scenes denoted their crucial roles in decoding the secrets in the whole movie. As well, the film has Oedipal and Eden-like overtones. Further, Cisse depicted the film’s Bambara natives and magical powers by means of an Albino and a dog walking backward (Diwara, 1988).Moreove r, a Hollywood movie with a character similar to Yeelen can hardly be found. Cisse tried to reveal the non-linearity of the Bambara’s belief; their religious belief is always moving back to the initial â€Å"brightness† that created the world. Transformational Leader Profile Essay One of the most debated, described and defined aspects of managerial competence, leadership continues to claim the attention of practitioners and theorists alike. Leadership, essentially a synthesis of arts, reflects individual experience, understanding, values and capabilities, interacting with situations where, realistically, there is rarely an ideal solution. The recognition that transforming a business is something of an heroic journey explicitly and implicitly acknowledges the key role of leadership in todays world as a determinant of success, in an organization’s change, progression and development. The quality of that leadership is itself largely determined by the leaders effectiveness as a coach and mentor who can grow the skills and confidence in others, necessary to shape tomorrows businesses, organizations and successes. To develop leadership one must work at developing himself.   The transformation of the self is central to the leadership development process. Meeting Peter was a very profound experience for me. I met Peter, CEO of a large healthcare management organization, a year ago. The CEO had recently been picked by the Chairman of the Board to head up the organization of approximately 20,000 employees. Everyone knew him, and it appeared that he was well-liked by employees and generally trusted. Peter is someone I totally admire. He is someone who always makes sure employees all know why they are part of the organization. The terms charismatic and transformational are used more or less interchangeably in much of the literature. Distilling a large literature on the transformational leader, the notion can be broadly captured by reference to six elements: an heroic figure (usually with attributed past success stories); a mystic in touch with higher truths; a value-driven individual rather than one who is apparently purely self-serving; someone who is perceived to know the way; an individual who has a vision of a more desirable and achievable future; and finally someone thought to be capable of caring for and developing followers. All six points reflect attributes of personality and behavior of Peter. Peter has an approach which aspires to significant organizational change through engaged and committed followers. It was John McGregor Burns (1978) who emphasized the meaning and significance of transformational leadership by contrasting it with transactional leadership (Orlikoff 2000). This theme was picked up and elaborated by Bass (1990). According to Bass, transformational leadership has four components: individualized consideration (the leader is alert to the needs of followers and also takes care to develop them); intellectual stimulation (the leader encourages followers to think in creative ways and to propose innovative ideas); inspirational motivation (energizing followers to achieve extraordinary things); idealized influence (offers followers a role model). The core of Peter’s model is cantered on the concept of developing oneself to develop others. Specifically, as Peter matures and gains moral perspective, he invests more time and energy in promoting the development of others versus satisfying his own needs. As Avolio and Yammarino (2002) note, through the accumulation of developmental experiences the moral structure of an individual can be enhanced providing sufficient structure to assess complex moral challenges. This basic premise is at the core of what drives transformational leadership to the highest end of the full range of leadership. In his daily work, Peter identifies developmental opportunities, where there is scope for: Challenge, and the breaking of new ground. Work that makes a significant, demonstrable contribution to the business. Bigger/wider leadership roles, preferably the earlier the better within the leaders career progression. Shifting gear and moving up into more strategic roles/tasks and relationships. Moving and operating out of existing comfort zones. Multi-disciplinary and cross-functional working. Building stakeholder coalitions and alliances, supply chain projects, involving suppliers, deliverers (own organization) and customers. Acquiring new contributive competencies which will test and enhance both skill and will. Working on specific role/task assignments in other comparable, but noncompetitive organizations. Making keynote presentations at major events, where there is a sense of occasion and opportunities to establish reputation and credibility. Leading (preferably) or acting as a member of jointly run projects with a leading business school, professional body or significant consultancy group. Setting up and managing increased outsourcing for supportive activities. In effect, leader learning development (like so much interpersonal skills training) is about building competence – and confidence – in three related areas of activity. On the reasonable assumption that competence – like charity – begins at home, the first priority in Peter development is learning to manage oneself. This includes not only the development of effective self-management skills, but also acquiring high levels of competence in the three primary areas of know-how identified as: Transforming Integrating Mobilising Peter’s personal skills and leader competencies form an appropriately strong base for developing consistently sound working relationships with others. Particularly important are the following interpersonal competencies: Self awareness and awareness of others. Negotiation techniques. Listening and questioning skills. Influencing skills. Coaching skills. Assertiveness. Motivational skills. Working as a partner (primus inter pares) as opposed to leader. Collaborative problem solving. Empowering and delegating skills. I identified five significant distinguishing characteristics which differentiated Peter from the less effective: He is neither perfect, nor perfectionists in their demands of others. He quickly learns skills which he does not possess personally. He sees management as essentially a team effort. He strives endlessly for improvement – to him, the game is not over until its over. He admits his weaknesses and learns from his mistakes, but differentiates himself from the less successful. The leadership theory proposed by Peter is based on the relationship and interaction between the leader and the follower. Under transformational theory, the leader and the follower may possess their own motivations for the interaction, but together they realize a common goal and are changed by the process. The change, or transformation, in the participants is found in the unity of purpose that raises the goal beyond individual satisfaction to a higher level. Peter’s theory is consistent with transformational theory. Collaboration cannot be achieved without the team members reaching a consensus and perhaps giving up something of themselves and changing in the process. Collaboration cannot be realized without facilitation, communication, information, participation, and expectation. Understanding something of the dynamics of group behaviour and the processes underlying transactions between people, is fundamental to effective leadership (Orlikoff 2000). There are no panaceas or cure-alls, but informed awareness of what is really going on, in behavioral terms, within and between groups helps to give managers – and others – a clearer idea of how to handle relationships in more productive ways. Peter believes that a business needs at least three interrelated forms of leadership: One which brings about requisite task performance and goal achievement. One to generate and maintain the commitment of its members. Another to ensure continuity of congruence between the requirements of the task, and peoples needs and expectations (Schein 1985). Peter has ability to overcome or compensate for (transform) organizational and individual limitations. He motivates others to do more than they originally intended and indeed often more than they thought possible. Team spirit is aroused. Enthusiasm and optimism are displayed. Peter enables his staff to overcome, to break through, to see beyond the limitations of their organization: he stimulates his ‘followers efforts to be innovative and creative by questioning assumptions, re-framing problems, and approaching old situations in new ways’ (Ackoff 1999). Peter is unique leader, on whom organizational success depends (and who therefore receives a considerable share of the benefits of that success). Peter seeks power not for self-aggrandizement but in order to share it. He empowers others to take an active role in carrying out the value-based mission or vision defined by him. That vision is based on what the organization and followers need, not what the leader wants personally. Thus, Peter appeals to followers values, emphasizing that certain important values serve as the common basis for our ideals and goals. Peter transforms organization by first using his cognitive power to understand complex causal chains and then acting to design outcomes that will benefit the organization and advance his vision. While a substantial degree of cognitive power is required in order for Peter to be effective, such effectiveness results as much from his success in developing followers cognitive abilities as from the exercise of his own. Peter with the degree of cognitive power required for a top-level position makes important long-term strategic decisions. But how much do these decisions affect what actually goes on in the organization on a daily, weekly, monthly, and yearly basis? It is the thought and action of managers and employees at lower levels that most affect current and short-term future operations. The finest long-term plan and the wisest long-range actions will surely fail if those who must act today and tomorrow are not capable of doing so. Thus, it is more important for top-level leaders with great cognitive power or vision to help followers expand and improve on their own vision than it is for leaders to simply exercise their cognitive power (Smith 2000). Peter empowers others to define organizational policies and develop programs that are explicitly based on the values and beliefs contained in the philosophy that in fact put those values and beliefs into organizational action. For example, hiring and promotion policies should take into account values consistent with those in the organizations philosophy as well as applicants knowledge and skill. Reward systems and bonus programs must be based on the values of cooperation and innovative action instead of on competition over a limited pool of resources. Finally, Peter inculcates values and beliefs through his own individual behavior, his personal practices. He models organizational values and beliefs by living by them constantly and consistently. That is why his leadership behaviors that were described earlier are extremely important. Many people think of these behaviors as tools with which leaders explain their vision to followers and convince them to carry out that vision. Although this is not totally untrue, the far more significant reason these behaviors are important is that leaders use them to demonstrate and illustrate the values and beliefs on which their visions are founded. Thats why Peter takes so much time and effort and why he is good manager with strong management skills. He uses everyday managerial activities a committee meeting, for example as opportunities to inculcate values. In a meeting the leader may guide a decision-making process while making it clear that final authority and responsibility rests with the group. By so doing, Peter takes what might otherwise be a bureaucratic process and instills the value of empowerment into it. Whenever possible, he overlays value-inculcating actions on ordinary bureaucratic management activities. Without a sound base of management skills, this would not be possible. Ultimately, examination of Peter’s leadership leads to the recognition that transformational leaders own personal behaviors play a large part in shaping organizational culture. This comprehensive theory goes beyond behavior to incorporate personal characteristics. Even more, it includes the organizational context of transformational leadership that is, culture building. Peter refers to the challenges as trigger events in our lives that oftentimes have a profoundly positive effect on our development. So how do we create the challenges that you must confront to develop into the full person you can be and to achieve your full potential? I am still searching for that in myself, and I hope you will do the same each and every day, because that is the way to develop leadership—each and every day we emerge, we get better, we know more, and we can influence people more effectively. Peter had a profoundly positive impact on my leadership development. I have chosen a developmental goal for myself, based on my primary style of leadership. After having examined Peter as a transformational leader I have tried to capture the whole process of personal and leadership development in a simple model. Let me explain. The top left-hand part of the model represents what we come into the world with our talents and strengths. Building on those capacities or attempting in some cases to break them down, we have life experiences that shape our development, that comprise our life stream. On the bottom left, we have the context in which we are currently operating and there we specify the importance of the vision and culture to nurturing leadership development. The rest of the model represents what we typically focus on in terms of leadership development, including enhancing our self-awareness of where we are and where we should focus our energies, then focusing those energies by regulating our development and then finally being consistent in our efforts to call it self-development. Ouchterlony Double Diffusion Assay Biology Essay Introduction: Polyclonal antibodies are produced by different B- lymphocytes in response to the same antigen, which recognise different parts of the antigen. Because the human immune system cannot know in advance what pathogens it will confront, it prepares for future infections by creating millions of different antibodies. Each of these highly selective proteins recognizes and binds to a specific target, or antigen, then signals other components of the immune system to destroy the target. These naturally-occurring polyclonal antibodies play a crucial role in triggering an immune response Polyclonal antibodies are routinely used as ligands for the preparation of immunoaffinity columns labeling reagents for the qualitative and quantitative determination of molecules in a variety of assays, such as double diffusion, radial immuno-diffusion, ammonium sulphate precipitation and ion exchange chromatography. Aim: The aim of this practical is to compare the purification of serum IgG by ammonium sulphate and ion exchange chromatography. The first purification step will normally involve a method such as fractional precipitation with increasing concentrations of ammonium sulphate. This method is not designed to achieve total purification, but to remove as much contaminant protein as possible whilst retaining all the protein of interest. Most proteins will precipitate from solution at high salt concentrations, but the salt concentration required to precipitate them varies considerably. Ammonium sulphate will be used as it is possible to set up salt concentration which will differentially precipitate serum proteins. Ammonium sulphate precipitation procedure was carried out to separate the serum proteins into four fractions. A fraction containing the serum protein to be purified can then be precipitated and collected, leaving behind any protein which is still soluble. The second method of purifying the IgG serum protein is ion exchange chromatography. This is a widely applied method of protein purification and uses positively charged groups or negatively charged groups immobilised onto a hydrophilic support, in this case DE- 52. Serum Proteins with an opposite net charge to that of the immobilised exchanger will bind to the column. Other serum proteins will pass through. Because the charge on proteins changes with pH, it is possible to attach a protein to the exchanger at one pH, then elute it by changing the buffer. Alternatively proteins can be eluted by passing an increasing concentration of salt through the column. The method works best for IgG which have high isoelectric points, at about pH 8.6. This method can also be used how to separate different subclasses of IgG. Ammonium sulphate is less effective in the purification if IgG, but it is useful for the isolation of large IgM. Samples of each fraction will then be separated by electrophoresis on an agarose gel. Antibody will then be allowed to diffuse towards the electrophoresed proteins from a trough cut parallel to the direction of electrophoresis. The proteins also diffuse from the positions they have reached after electrophoresis and precipitin arcs form where antigen and antibody reach equivalent concentrations. This technique can be used to determine whether a fraction contains any IgG and determine the degree of contamination of the IgG with other proteins Materials and Method: Ammonium Sulphate fractionation Procedure 0.25 ml of saturated ammonium was added to 1ml of human serum, to produce a solution which 20% saturated with respect to ammonium sulphate. The solution was mixed it was allowed to stand in an ice for 15 minutes, and was centrifuged for 15 minutes at 1500 rotation per minute. The supernatant was poured and pallet was retained as fraction 1 The supernatant from fraction 1; 0.35 was added to bring to 35 % saturated with respect to ammonium sulphate; the solution was left in an ice for 15minutes and it was centrifuged to recover the precipitate, the supernatant was poured in another tube while the pallet was retained as fraction 2 0.5 ml of saturated ammonium was added to the supernatant of fraction 2 to bring the solution to 50% , the solution was left in ice for 15 minutes to precipitate, it was centrifuged for 15 minutes the pallet was kept as fraction 3 while the supernatant containing 50% of protein was kept as fraction 4 the absorbance of the fraction was measured at 280nm the absorbance of 1mg/ml and 0.5mg of bovine serum albumin was measured was measured Before the immunological analysis the fraction salt content were reduced by dialysis against buffer. 0.2 (For more information refer to UEL hand out on protein purification) DE 52 ion exchange chromatography Serum provides was pre dialysed against 10mM trs/barbitone buffer pH 8.6 and chromatography column containing about 2mls of DE 52 which it has been equilibrated in the same buffer The column was allowed to run until any overlying buffer has run into the DE-52 gel avoiding the column to dry Ouchterlony Double Diffusion Assay The fractions collected from ion exchange chromatography were determined for the presence of IgG by using ouchterlony double diffusion method. The collected fractions were run against an anti-IgG antibody in an agarose gel. The centre well were filled with 3ul of anti-IgG and 3ul of the eluted fractions into the surrounding holes. Immunodiffusion was slowed to proceed for 24-48 hours an antigen-antibody precipitin line was observed. Single Radial immune diffusion This as a quantitative technique whereby the antigen is allowed diffuse from a well into a gel which contained its specific antibody, a precipitin will form when antigen concentration is equal to the concentration of the antibody in the gel. Immunoelectrophoresis MATERIALS AND METHODS Preparation of antigen Blood samples were collected from ten clinically healthy cows using sterile disposable needles (1.2 40 mm), clarified by centrifugation (1000 g, 15 min) and diluted 1:1 with phosphate buffer saline (PBS, pH 7.2). Then equal volumes of diluted serum and saturated ammonium sulphate were mixed by slowly addition of the saturated ammonium sulphate solution with gentle stirring. After centrifugation (1000 g for 20 min), the precipitate was washed twice with 50% saturated ammonium sulphate solution. The final precipitate was dissolved in PBS followed by overnight dialysis against PBS. Protein concentration was quantified by a coomassie dye binding assay (Bradford, 1976), using bovine serum albumin (BSA) as the standard. Final protein concentration of solution adjusted to 1 mg/mL. Immunization of rabbits with bovine immunoglobulins Three hundred micro liters of prepared bovine immunoglobulins (1 mg/mL) in PBS was emulsified with equal volumes of Freunds complete adjuvant (Sigma) and inoculated intramuscularly (I M) into three 6-month-old New Zealand White rabbits. The rabbits were fed regular commercial diets. The second and third inoculations were performed on days 21 and 35 with Freunds incomplete adjuvant (Sigma), and the fourth inoculation was done on day 45 without any adjuvant. After the final immunization, blood samples were taken from the rabbits and production of antibody was investigated by double diffusion and ELISA tests. This study was approved by the Regional Medical Sciences Research Ethics Committee of Tabriz University of Medical Sciences. Purification of rabbit anti-bovine immunoglobulins Immunized rabbits sera were collected and precipitated by 50% ammonium sulfate. After dialysis against PBS and tris-Phosphate buffer (40 tris and 25 mM phosphate, pH 8.2), ion-exchange chromatography was done on a DEAE-Sepharose fast flow (Pharmacia) in a laboratory made column at a flow rate of 0.25 mL/min. Protein concentration adjusted to 100 mg/mL and passed through the column. The column was washed in two steps using Tris- Phosphate buffer for first washing step and Tris-phosphate buffer containing100 mM NaCl for second washing step. The eluted proteins were collected in 5 mL fractions and analyzed by SDSPAGE. SDS-PAGE analysis The purity of various IgG preparations was checked using sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) under reduced conditions as described by Laemmli (Laemmli, 1970). The final concentration of polyacrylamide solution was 13%. Samples were boiled with 2% SDS for 10 min and were loaded on the electrophoresis gel. After separation, the proteins were stained with Coomassie Brilliant Blue G 250 (Blakesley and Boezi, 1977). Destaining was carried out in distilled water. Conjugation of rabbit IgG with peroxidase The conjugation was performed by the periodate method (Nakane and Kawaoi, 1974) with some modifications. First, 4 mg of peroxidase (Sigma) was dissolved in 0.5 mL of distilled water in darkglass container. Then sodium periodate (Merck) was added to the solution, and the container was kept on a stirrer for 20 min at room tempe-rature. The mixture was dialyzed against acetate buffer (0.1 mM, pH 4.4) at 4Â °C overnight followed by addition of 10 ÃŽ ¼l of carbonate-bicarbonate buffer (0.2 M, pH 9.5). Eight milligrams of purified IgG in 1 mL of carbonate-bicarbonate buffer (10 mM, pH 9.5) was add-ed to the active enzyme, and the container was put on the stirrer. Then 150 ÃŽ ¼l of fresh sodium borohydrate solution (Merck) was added to the above solution and was kept at 4Â °C for 1.5 h on the stirrer. The product was then dialyzed overnight against PBS at 4Â °C and 1% BSA (Sigma) along with addition of 0.01% sodium mirth-iolate (Merck). Enzyme linked immunosorbent assay (ELISA) Direct ELISA was used to determine the titer of HRP conjugated rabbit IgG against bovine immunoglobulins. 100 ÃŽ ¼l of prepared bovine, sheep and goat immunoglobulins, which was diluted 1:100 in PBS (10 ÃŽ ¼g), was added to each well of a 96-well micro titer plate and incubated at 4Â °C for 24 h. The wells were washed with PBSTween (0.05% Tween 20) three times and blocked with 200 ÃŽ ¼l of blocking solution (PBS-0.5% Tween 20). After a washing step, 100 ÃŽ ¼l of 1:400, 1:800, 1:1600, 1:3200, 1:6400 and 1:12800 dilutions of prepared HRP conjugated anti-bovine immunoglobulins were added to each well. The reaction was developed using 100 ÃŽ ¼l of 3, 3, 5, 5- tetramethylbenzidine (TMB) as substrate and the absorbance was determined at 450 nm after stopping the reaction by 5% sulfuric acid (Sigma). Results: RESULTS Production of rabbit anti-bovine immunoglobulins In order to survey production of antibody in rabbits and evaluating effectiveness of immunization, double diffusion and ELISA tests were performed. The titer of polyclonal anti-bovine IgG in double diffusion test was 8, which appeared as a sharp band between antigen and antibody wells. The titer of anti-bovine immunoglobulins determined by ELISA was 16000. Purification of rabbit anti-bovine immunoglobulins Purification of IgG rich fraction from immunized rabbit sera by ammonium sulfate precipitation followed by DEAE ion-exchange chromatography resulted in a highly pure fraction (first peak). The protein content of this fraction was 45 mg which was about one third of primary protein content (Figure 1). SDS-PAGE analysis Figure 2 shows the results of SDS-PAGE for determining the purity of IgG, which was purified by ion-exchange chromatography. A distinct polypeptide band with molecular weight about 50 kDa corresponding to rabbit IgG heavy chains. The diffused bands between molecular weights of 20 30 kDa correspond to rabbit IgG light chains. (Figure 2) The SDS-PAGE analysis showed that purification of IgG by ion-exchange chromatography resulted in a highly pure product. Discussion: The purification of immunoglobulins presents several practical complications, especially for polyclonal antibody production (Verdoliva et al., 2000). We used ionexchange chromatography for purification of rabbit IgG polyclonal antibody. Separation and recovery of proteins from ion exchange chromatographic media are affected by factors such as buffer type and pH, length of gradient, flow rate of the mobile phase, ionic strength and nature of counter ion, and characteristic of the proteins. The selection of ideal conditions for protein purification involves changing some or all of these parameters (Tishchenko et al., 1998). This technique was well established in our laboratory for purification of IgG antibody (Baradaran et al., 2006; Javanmard et al., 2005; Majidi et al., 2005). Furthermore, ion-exchange chromatography is considered as an economical alternative to affinity and immunoaffinity chromatography. After purification step we obtained a protein with approximate purity of 98%. SDS-PAGE analysis showed that the protein with approximately 50 kDa MW was rabbit IgG heavy chains. The light chain of rabbit IgG appeared as a diffused band of 20 30 kDa molecular weights. It is likely that diffused band of light chain could be related to different level of deglycosilation of protein during manipulation process. Conclusion:

Life and Times of Thomas Day   Ã‚  Ã‚  Ã‚  Ã‚  Thomas Day was born in Dinwidee, Virginia in 1801 to a free slave mother. With the law that allowed children to be born free if their parents were free, Day was born free and did not have to be a slave. His family had been free since the early 18th century. He and his brother were educated by private tutors and they were trained by their father in cabinetry and carpentry. Thomas’ brother eventually began to study theology and he emigrated to Liberia in 1930 and was a Baptist missionary, eventually became one of the signers of the Liberian Declaration of Independence and a prominent statesman there. He’s known as one of the founding fathers of that nation.   Ã‚  Ã‚  Ã‚  Ã‚  Thomas moved to Milton, NC in 1823 and started a furniture business where he became one of the best furniture makers of that time. He received notice from two of North Carolina’s governors and his furniture is inside of University of North Carolina in Chapel Hill’s original buildings. He eventually began to train free blacks and enslaved blacks to do the carpentry work that he was doing. By the mid 1800s his work was in demand from Virginia to Georgia. He then had to begin purchasing slaves to help him to the work. His furniture shop became the biggest shop in the state because he used steam powered machines and using mass production techniques to build his furniture.   Ã‚  Ã‚  Ã‚  Ã‚  In 1827 Thomas married a free woman from North Carolina. But the law was that free slaves could only marry people from their own state. So Thomas threatened to move his furniture shop outside of North Carolina and to move it to Virginia where that law did not exist. So the North Carolina Legislature made special arrangements to the law so that Thomas would stay in North Carolina and he and his wife could still be married. Thomas had three children and they were educated in an abolitionist-sympathizing school in Massachusetts called Wesleyan Academy. He also constructed some pews for The Presbyterian Church in Milton provided that his family would be allowed to sit up front in the white section. He was an active member of the community of Milton. In 1848 he purchased the Union Tavern where he resided until he died in 1861. It is now a landmark but part of it was destroyed by a fire in 1989.

Efficacy of Portable Head CT Scan in the Neuroscience Intensive Care Un

Introduction: Portable Computed Tomography (CT) studies are an important component used to manage, assess and diagnose CNS diseases, and acute brain injuries, in the Neuroscience ICU (NICU). The portable device is designed specifically for head and neck scans for critically ill patients that are at risk for complications and increased morbidity during intrahospital transportation. Research has found evidence which substantiates that intrahospital transport of patients with brain injuries can impact their outcomes. Many hospital protocols dictate the use of portable head CT (pHCT) scanners to monitor and assess critically ill patients in the NICU, to decrease negative patient outcomes from intrahospital transportation. This is an important factor in reduction and prevention secondary injuries in critically ill patients. The intention of this paper is to critically analyze a research article entitled Portable Head CT Scan and its Effect on Intracranial Pressure (ICP), Cerebral Perfusion Pressure (CPP), and Brain Oxygen. First, there will be an article synopsis identifying the premise of the study. Second, validity of the study will be described and discussed. Lastly, this paper will discuss applicability to Neuroscience ICU at University of New Mexico (UNM) Hospital. Article Synopsis: The authors of this research article were from various Departments at University of Pennsylvania in Philadelphia. The researchers conducted a retrospective study to assess possible detrimental association of a pHCT on ICP, CPP, and brain oxygen in patients with severe brain injury. The retrospective study selected 34 patients (16 males and 18 females), with the mean age of 42 ( ±15 years), and Glasgow Coma Scores (GCS) ≠¤ 8 at the time of pHCT sca... ...eterious effects on patients. Conclusion: Peace et.al. raises a valid hypothesis that warrants further investigation in order to decrease the risks to patients on NICU and other units/ hospitals. In my opinion, studies with significant findings, reliability, high internal and external validity, are imperative in making changes in hospitals around the world to decrease secondary injury to patients. This study can possibly entice other researchers to develop a better study design and investigate the relationship between pHCT scans and ICP, CPP and brain oxygen, more vigorously. Introduction Management and leadership skills in nursing and other healthcare professions are becoming a more and more widely spoken of subject, as they are essential skills for everyday practice in delivering care (Gopee and Galloway, 2009). Effective management requires the nurse to be able to draw on evidence based knowledge and experience to develop the ability to manage competently during practice. Skills without knowledge, appropriate attitude and understanding will not equate to competency (Watson, 2002). This piece of work will critically explore the theories about management and leadership in health care. At the beginning it will provide a base understanding of both topics and accountability importance. It will then concentrate on management of conflict, which could be seen in the clinical setting scenario from my current placement area. It will demonstrate how the use of conflict management theories can be helpful and how I behaved in the role of a manager. Also different leadership styles involved during this conflict situation will be analysed with use of evidence-based literature. To address the subject of confidentiality as per the Nursing & Midwifery Council guidelines (NMC, 2008), no names or exact location of the hospital placement will be mentioned. I have developed knowledge about conflict management theory, especially the importance of trying to achieve a win-win situation between the two different leadership styles, to be precise, autocratic and democratic. Professor Dr. Muhammad Yunus Muhammad Yunus is a banker to the poor and economist who was born on 28 June 1940 in Bangladesh. He was a professor of economics and is famous for his work in microcredit. Professor Muhammad Yunus established the Grameen Bank in Bangladesh in 1983 to help the poor by giving out small loans. He turned the idea into  Grameen Bank, which has made small loans to more than 7. 5 million people now. His first loan was $27 given from his own pocket. This $27 was loaned to 42 women in the village, who made a profit of $0. 2 each from the loan. The activity of the Grameen Bank is known around the world. Its ideas have been used throughout the world including USA, China, Malaysia, India etc. More than 94% of these loans have been given to women to improve their lives and escape poverty. The Norwegian Nobel Committee decided to award the Nobel Peace Prize in 2006, divided into two equal parts, to  Muhammad Yunus  and  Grameen Bank  Ã¢â‚¬Å"for their efforts to create economic and social development from below†.Muhammad Yunus has received several other awards. Microcredit means giving small loans. These loans are given to poor people. To qualify for a loan, the villager must demonstrate that their family owns less than one half acre of land. The activity of Grameen Bank has not been limited to just Bangladesh but also around the world. Grameen helps the world’s poorest, especially women, improve their lives and escape poverty. More than 94% of Grameen loans have been given to women, who have less money and give more to their families.Grameen Bank was the first lender to hand out microcredit, giving very small loans to poor Bangladeshis who did not qualify for loans from normal banks. The bank has provided $5. 72 billion dollars to over 6 million families in Bangladesh. These loans average $200. With 1,417 branches, Grameen provides services in 51,000 villages, covering three quarters of all the villages in Bangladesh. This system was so successful that even beggars had been able to borrow money.The people who have enough money has many banks to support them, but those who don’t have anything, traditional banking system, that’s why Muhammad Yunus created the Grameen Bank and thus winning the Nobel Prize for Peace. The main aim of the traditional bank is to provide loan in exchange of depositing the property or other assets with interest. Paying Division One Athletes

ENG 100P April 12, 2013 Pay to Play: Should Division One College Athletes be Paid? Every year division one college athletes put everything they have on the line during practice everyday and make hundreds of millions of dollars for their schools and the NCAA every year. These athletes aren’t just playing for the love of the game anymore once they reach the division one level, it turns into more of a job than an activity. The players spend hours every single day dedicating almost all of their time to the sport they play.These athletes work just as long and much harder than your average person working a nine to five job, so why don’t the athletes get paid for their commitments to their teams? Last year the NCAA made 871. 6 million dollars from division one sports. The average division one men’s football team brings in 15. 8 million dollars to their school each year. The amount of money that these sports make for their school is so high; the players should get some k ind of reward for all they do for their schools.The old rules that say college athletes cant be paid in any way needs to change, division one college athletes deserve to be paid for all they do for the NCAA and their schools. Growing up every athlete’s dreams of playing in that national championship game for their dream school and scoring the winning touchdown or overtime goal. As a child you don’t realize that commitment it takes to be part of a division one athletic team. Coming from a big lacrosse and football town, my friends and I have gone through the division one recruiting process and know how hard it is to play a division one sport.Many of my former teammates play division one lacrosse and after talking to them I realized really how tough it is to play a division one sport. When asked about how lacrosse at Sacred Heart University was going, Freshman Spencer Hackett said; â€Å" I’ve grown up with lacrosse and I love the sport very much, but division on e is so much more then I thought it would be. Everyday I have two practices that last at least two hour, then after that I have mandatory lift and film sessions. Spencer dedicates at least six hours every day for his team, and they’re one of the lowest ranked teams in division one, I can only imagine the commitment it takes to play on a high ranked team. In my personal experience of going through the division one recruiting process, I saw how much blood; sweat and tears go into being a division one athlete. On my overnight visit at University of Massachusetts, which is a top 20 team, most of my day was watching the team practice, or lift, or watch film.It seemed like everything they did was revolved around lacrosse and to me that’s not what college is about. These division one athletes have such little time to socialize and make friends outside of their sports; everything they do is revolved around their sports. The fact that athletes willingly dedicate so much time to their sports is a huge reason why they should be paid. Division one athletics, especially sports such as football and basketball, are extremely profitable for the NCAA and for their schools. According to NCAA. org, the NCAA’s revenue for the 2011-2012 athletic seasons was 871. million dollars. The NCAA makes an extremely large amount of money from division athletics, with most of the money coming from television contracts and championships such as March Madness. Schools such as Texas make hundreds of millions of dollars because of their excellence in many sports, last year their football team profited $68,830,484. A lot of the money schools make, such as Texas, is from selling merchandise. Big name schools sell tons of merchandise to the public; selling products such as jerseys should be illegal if they players don’t get paid.Why could they sell jerseys with players number on them but that player doesn’t get compensated for using their number on the jersey? Rob ert and Amy McCormick of Michigan State University have added a new dimension to the long debate over paying athletes by arguing they are â€Å"employees† under federal labor laws and entitled to form unions and negotiate wages, hours and working conditions. † Robert McCormick was the former attorney for the Nation Labor Relations Board and if he is saying that these players are employees, it’s about time that the NCAA recognizes them as employees also.A common argument against paying college athletes is that they won’t compete as hard because it won’t be the same as when they weren’t getting paid, but that argument has absolutely nothing to do with paying players. If anything, paying the players would make them work harder so that they could make more money! I believe that the best way to go about paying these players would be contracts, just like the professional athletes and like any other employee in the United States.These contracts woul d go on to say that the players would be paid an amount depending on their contribution to the team and that students would need to finish their schooling so that they have a backup plan. That is why you’re supposed to go to college in the first place, isn’t it? So that you can get a good education! To do this the NCAA would need the support from the professional sport teams and not draft players who haven’t graduated college yet. Another argument why players shouldn’t be paid to play is because they students-athletes and that there is a reason student becomes before athletes.Realistically, these young men and women aren’t student-athletes at all. Many of the players on division one team would never have gotten into college if it weren’t for sports. Also, if they are student-athletes then where do they find the time to do their schoolwork when they have practice, film, lifting, running etc. all day? The only reason many athletes go to colleg e is so that they can get looks from the pros. If they are student-athletes then why do many of them leave college after a year or two to play professional sport?It’s because college is just a stepping-stone to them, they don’t care about the school aspect. When it comes to division one athletes, they are athletes first and students second. The players know it, the coaches know it, the fans know it, the only people who don’t know it are the people who make up the NCAA. In conclusion, times have changed and it’s time to pay division one athletes for their commitment and for the money they bring in for the schools and the NCAA. With college athletics competition being so tough now and so competitive, players have to dedicate so much more time to their sports then in the past.College athletics are not longer a sport that players do for fun, college athletics are now a job for the players that consists of hours of practice everyday and throwing your social li fe out for your sports college athletics make hundreds of millions of dollars for the NCAA and tens of millions of dollars for the schools, so why is it that these players don’t get compensated for their commitment and contribution to the schools and the NCAA? I personally believe that it is ridiculous that players don’t get paid anymore.One of my best friends Dylan Baumgardner, a lacrosse player at Quinnipiac University said it perfectly when asked if he still loved playing lacrosse, â€Å" I don’t play lacrosse anymore, I go to class, then I go to work all day, then I go to sleep. † Joe Nocera of the New York Times, came up with a plan for college athletics that would make college athletics work like professional sports with signing bonuses, salary caps, insurance, player unions and it would even offer additional scholarships to players who want to further their education. This plan will go into consideration in 2014.
